1:Recent developments in modeling HIV infection and treatment

Highly active antiretroviral therapy can suppress viral replication but cannot eradicate the virus. Mathematical models, combined with experimental data, have provided important insights into HIV dynamics, immune responses, and drug treatment. However, there are still a lot of questions that remain unanswered, for example, are some drugs more effective than others? whether treatment intensification brings benefits? what causes multiple infection that may lead to drug resistance and immune escape? are there any other sources, besides the latent infection, that contribute to HIV persistence despite long-term therapy? In this lecture, I will present some recent modeling efforts in addressing these issues and also discuss their implications for the management of HIV infection.

2:Modeling HCV dynamics

Traditional therapy with interferon is not effective in treating chronic hepatitis C virus (HCV) infection. New treatment using direct-acting antiviral agents (DAAs) has significantly increased the cure rate. These new DAA drugs directly interfere with different steps of the HCV life cycle. Thus, the classical viral dynamic models that do not consider intracellular processes may not be optimal in analyzing data from patients treated with these drugs. In this lecture, I will discuss some advances in developing multi-scale mathematical models that can study HCV dynamics under therapy with new drugs. I will address model analysis, approximation, comparison with experimental data, as well as the implications for developing new treatment strategies for hepatitis C.

3Within-host dynamics of influenza virus infection

Influenza virus infection remains a public health problem worldwide. The mechanisms underlying viral control within an individual with an uncomplicated influenza virus infection are not fully understood. In this lecture, I will address this by reviewing existing work and presenting a model including immune responses. I will discuss the relative roles of target cell availability, and innate and adaptive immune responses in controlling the virus. I will also discuss combination therapy for influenza.
